I don't understand why it would regenerate inner hair cells and not outer. The whole purpose is to "activate" progenitor (supporting) cells in the cochlea, whether inner or outer. It seems these guys have no idea what they are doing. I hope I am wrong though.

Imagine FX-322 or FX-345 actually restores hearing but since the benefits are not long lasting, you suddenly lose your hearing on an important day such as work. LOL. That would be funny.

I believe they are taking a risk by focusing on repair of hair cells. Since many people with significant / severe hearing loss could have either damage or hair cell death. And if patients with hair cell death participate in their trials, this drug will definitely fail.

It would be better to wait until Phase 1b results are out in the first half of 2023. And if results are positive (better than FX-322), maybe they will allow Expanded Access? Let's hope for the best.

Does FX-322 restore hair cells only? I read somewhere it's not only for hair cells but cochlear synapses as well.

Well, if the hair cells are weaker, then they should stop this PCA nonsense and let the grand daddy of inner ear disorders (which is Otonomy) do the job for them! We don't want weak hair cells!

Can someone please explain how the 5 subjects in the image above started to lose the benefits from FX-322 after only a couple of years? From what I know, it takes tens of years for someone to start losing hair cells even if exposed to high levels of noise. Can anyone intelligent explain?

I don't think it will take that long (2030) for FX-345 to be in the market. They are going to start the Phase 1b trial this year and will end in the first half of 2023. Phase 2 trial also takes a year. While Phase 3 could take up to 2 years. So maybe in like 4 years (2026) we might get our hands...

Does this drug restore hearing permanently or temporarily? I know that hair cells can be lost due to noise, aging, and other reasons but I can see from the corporate presentation that the gained hearing is lost after 1-2 years. Can someone explain why that is please? Is Frequency Therapeutics...

Even if the new hair cells are weaker, if I have to take this drug (in case it works) every 4 months, I would have no issue at all with that.

The exclusion criteria for OTO-413 is because some patients might not notice the improvement in hearing due to their LOUD tinnitus. Nothing more.

Only 6 dB improvement and they call it "clinically meaningful". Who is going to pay hundreds of dollars for a slight yet not even noticeable improvement in hearing?
