    Stem Cell Secretome Treatment

Thanks. It is not publicly available, this is my own making, in a laboratory. And yes, it is an intratympanic injection, injected cold, it gels when it reaches body temperature, hence the slow release. Modified FX-322 was not a miracle, maybe slight benefit to one ultra high frequency, also...

    Stem Cell Secretome Treatment

The exosomes that I receive are derived from umbilical cord mesenchymal stem cells. I can get the cells themselves, too, but cell therapy is riskier (and the cells will likely not cross the round window if injected intratympanically). I'll reiterate the same thing I have posted before...

Frequency Therapeutics — Hearing Loss Regeneration

Could you please provide any publications on intratympanic exosomes then? I'm talking human trials, I do know they seem to be effective in animals. However, hearing-wise, they did nothing to me (systemically at very high doses). I will try growth factors (BDNF and/or NT-3) intratympanically...

Frequency Therapeutics — Hearing Loss Regeneration

I personally know two people who have had stem cell exosomes administered intratympanically (1 week of IT shots, a shot every other day, so either 3 or 4 shots, I forget). No improvement. One shot was around 0.5 ml, containing 50-100 billion exosomes. All that aside, the PCA approach worked...

Otonomy OTO-413 — Treatment of Hidden Hearing Loss

For those interested, Spiral Therapeutics has patents on their slow release gel. It's a chemically crosslinked polymer - you mix PEG esters with trilysine amines and can control the crosslinking reaction by adjusting the pH. This gel is more durable than a P407 gel (Otonomy and Frequency...

Frequency Therapeutics — Hearing Loss Regeneration

To be honest, FX-322 did reach 10 kHz at required concentrations of CHIR99021 and sodium valproate to work. Now testing speech was an interesting choice, but it makes sense because understanding speech is one the main goals of hearing restoration. Getting deeper required *some* changes to the...

    Frequency Therapeutics — FX-345

It was not. The regular FX-322 is 88 mg/ml sodium valproate (HDACi), 3.1 mg/ml CHIR99021 (GSK3i), 5% w/v DMSO (solvent for CHIR) and 16.1% w/v Pluronic P407 (gelling agent). FX-345 is likely (based on very limited info they released) the same 88 mg/ml sodium valproate, 5% w/v DMSO and 16.1% w/v...

    Stem Cell Secretome Treatment

Not worth it, as the number of exosomes is low for the price. Even with really high doses IV (not their exosomes), I saw no improvement in hearing or tinnitus, but my hyperacusis is gone. Unsure if that was natural or that exosomes helped.

Visual snow is the same as it is likely directly correlated with the signal loss due to hearing loss. Again, I cannot recommend this treatment because of the decently high cost, the low absolute numbers of exosomes and the lack of evidence of efficacy.

It was both. At this point I would not recommend it, I think one needs to have a decently high concentration of growth factors and/or exosomes on the round window for this kind of stuff to be effective.

Hi, Still nothing. But I am hyperacusis-free, so that is good. I am continuing with intravenous umbilical cord stem cell exosomes, at very high doses (2 trillions every few weeks, compared to around 20 billion at the clinic). But so far nothing. The next step would be intratympanic things. :-)

I have no other conditions, except maybe some red spots on my face. The exosomes cleared it up a bit. Just to reiterate, in my opinion, their dosage is way too low for the price you pay. Will You Get Vaccinated for Coronavirus (COVID-19)? Can It Make Tinnitus or Hyperacusis Worse?

Will You Get Vaccinated for Coronavirus (COVID-19)? Can It Make Tinnitus or Hyperacusis Worse?

I got another Pfizer booster and I am completely fine. I had a sore arm for one day, that was it. Tinnitus is quiet. Get vaccinated because COVID-19 DOES hurt the auditory system. With the rate that Omicron is spreading, most people will have some contact with the virus, unless they can isolate...

I had ear pressure after exposure to sound, and if I continued exposing myself to sound, it would become painful - pain deep inside, sort of going into my throat. It is better now. But again I cannot say whether this is due to the exosomes, or simply time. Maybe, I am waiting, of course...

I am not the forum's favourite hydrocarbon, but I can try to answer this! Their new GSK3i is supposedly more potent (replacing CHIR99021) and they're possibly using a higher concentration of Valproic Acid, courtesy of using a purified Poloxamer P407 (unpurified P407 will not allow for higher...

    Stem Cell Secretome Treatment

I personally would not do stem cells, only exosomes/secretome. Simply because the cells will only get to your cochlea in small numbers and they are unlikely to engraft there. And also the added danger of injecting live cells (especially allogeneic) vs injecting small extracellular vesicles.

    Hyperbaric Oxygen Therapy

You are misunderstanding me, it's not about oxygen intake, it's because you are in an airtight capsule while you undergo the procedure and they increase the air pressure there; if you can't equilibrate pressures, you might get barotrauma.
  53. Tau

    Hyperbaric Oxygen Therapy

The vented plugs have a hole for the air to escape, thus making it easy to equilibrate the pressure between the outer ear and the middle ear. I used my musician's plugs.

I Invented a Sound That Knocked Out My Tinnitus

I Invented a Sound That Knocked Out My Tinnitus

This, I would like to add that unwinding a double-stranded DNA helix takes a lot of energy. Even if one base-pair disconnects due to thermal energy, it will reconnect because the others are still holding the helix together. For a 12 base pair DNA strand (very short!) to completely unwind, it...
