New University of Michigan Tinnitus Discovery — Signal Timing

Auricle is a dead horse now. Beyond science, Pearson's incompetence will make it explode. It's a succession of red flags without the slightest positive sign. Even if he manages to bring it to market in several years, there will likely be better treatments available by then, or the patent will...

Xenon Pharmaceuticals' XEN1101 — Kv7 Potassium Channel Modulator

In the pharmaceutical world, tinnitus doesn't seem to rhyme with "gold mine;" in my opinion, it's quite the opposite. Very few conditions are more cheesy and despised than tinnitus.

Xenon Pharmaceuticals' XEN1101 — Kv7 Potassium Channel Modulator

You forgot the story of people turning blue (irony). Trobalt was not the poison some people would have you believe. Definitely a drug with too many side effects, I agree. And XEN1101 will in all probability have side effects of the same nature, although attenuated.

I think that instead of making assumptions (positive or negative), it would be more helpful if everyone consulted Dr. Shore's patent.

Lenire — Bimodal Stimulation Treatment by Neuromod

In Dr. Shore's paper, timing is parameter #1. Lenire is based on a different theory I know, but where Dr. Shore's papers are well constructed, Lenire's are enigmatic. In my opinion, using a Bluetooth system in such sensitive device is an unnecessary source of complexity, to favor the...

Lenire — Bimodal Stimulation Treatment by Neuromod

Lenire uses a Bluetooth system, which is a disaster for a system where the timing must be precise to a millisecond range. This unnecessarily complicated design makes Lenire closer to a teleshopping gadget than a medical device. Comparing Lenire with real bimodal treatment devices makes no sense.

Oral Gabapentin and Intradermal Injection of Lidocaine: A Role for the Treatment of Tinnitus?

Of course it's your choice, but Lidocaine alone does not have much benefit beyond what we know. Personally, I have never had any problems with Gabapentin, even at high doses. Maybe consider low starting doses? It's a real chance to have such an understanding and competent ENT.

Is there evidence that the GABA activity of Retigabine is a different mechanism from other drugs that act on this neurotransmitter?

To put it simply, yes, I got an objective reduction with Retigabine, because I simply no longer heard my somatic tinnitus, like 0.

Xenon Pharmaceuticals' XEN1101 — Kv7 Potassium Channel Modulator

Our greatest fear is that the selectivity of XEN1101 doesn't activate a channel necessary for tinnitus suppression. I'm of course thinking of Kv7.4 which is present in the auditory pathway. Maybe I'm misinterpreting, but from what I understood from these two papers: XEN1101 Retigabine...

No, the stimulation is not more important than the sound. In fact, according to the papers, the stimulation part alone tends to temporarily worsen tinnitus. To put it simply, this technology consists of stimulating neurons via somatic stimulation and sound, almost simultaneously, but with a...

I guess it will take another year after the FDA decision to think about what to do, and waiting another year to start thinking about production after. I respect Dr. Shore's work but I come to think that the best way for her technology to come out, is to have it stolen by the Chinese who don't...

Oral Gabapentin and Intradermal Injection of Lidocaine: A Role for the Treatment of Tinnitus?

It would not be a surprise for me if a relatively effective path remained forgotten. I remember the total immobility of tinnitus sufferers during the Team Trobalt era. Gabapentin and Lidocaine represent some interest. But I don't really understand, it's an intradermal injection but in the...

Xenon Pharmaceuticals' XEN1101 — Kv7 Potassium Channel Modulator

Apart from the tolerance which can develop over time, like a lot of drugs, the random effects of Retigabine must be able to be explained in my opinion. This effect on smooth muscles may be the cause of certain side effects of Retigabine. Particularly on urinary retention and cardiac “disorders”. So, if I recall correctly, it's a channel that reformulations avoid. According to Professor Tzounopoulos, Kv7.2/3 are clearly the induction...

Reformulated Retigabine (Trobalt): BHV-7000, a Kv7.2 Modulator by Biohaven (Pfizer)

Interesting. This may be one of the explanations for why Retigabine doesn't work for everyone. But according to the anecdotes we have, it's mainly, in my opinion, a difference in generation site. A good % of people benefit from Retigabine, at least a little, which indicates that their Kv7...

These heavy side effects were also associated with Gabapentin and my emotional state so it's hard to be sure. I prefer to warn when in the doubt. It's also quite comparable to acids/hallucinogens, the risk of "bad trip" and psychosis is mainly linked to your emotional state. That's what I felt...

Yes, this has always been the case so far, from almost nothing to heavy side effects. I think it's related to CI in my case, bc this T can reach other side, at least not in brain. Idk at all, RTG is so random, i took time to make it work for somatic one, and non-somatic one seems to respond a little, so i still have hope.

It helped my somatic tinnitus, yes, it 90% vanished for 1 hour. It probably did something for the non-somatic tinnitus, but far less than for my somatic tinnitus. But I remember I was one day basically tinnitus-free for 1 hour, i.e. impossible to hear it unless being in perfectly silent room.

My somatic T generally responds well to RTG, can reach 90% at hight dose. My non-somatic T reacts only slightly (20% placebo range), but I think there is still an impact. This is my personal theory, I think that my non-somatic T is located in a structure superior to DCN, which responds less...

And some money consideration, to be honest. Again, Retigabine indeed has heavy side effects but these mainly concerned high doses in the long-term. It's not poison. A reformulation halving the side effects would be, in my opinion, already acceptable.

Xenon Pharmaceuticals' XEN1101 — Kv7 Potassium Channel Modulator

Dude, I'm not changing the subject at all. I don't know what you're trying to say, it's pretty well documented here that Kv7.2 and Kv7.3 are suspected to be the source of some cases of tinnitus, more than any other channel. But other mechanisms are possible of course, that's all my doubt...

Well, in most of the papers I've read about Kv7s and tinnitus, there's rarely any mention of channels other than Kv7.2 and Kv7.3. If you have papers which explore the application of other channels in tinnitus (and not in hair cells), I'm interested. For what it's worth, if I recall correctly...

Xenon Pharmaceuticals' XEN1101 — Kv7 Potassium Channel Modulator

That's exactly the point. I don't have an answer, I just know that according to the dominant theory Kv7.2 and Kv7.3 are "enough". K7.4 is indeed expressed in the hair cells. In my case I am 95% sure that the tinnitus affected by Trobalt is not located in the hair cells but in the DCN. It is...

In my case, Gabapentin is incredibly weaker in terms of efficacy on tinnitus than Retigabine (Trobalt), although I think they make a good combo. I used Gabapentin with a Kv7 opener. From what I understand in my case (related to hearing loss), tinnitus is largely a central problem, maybe even totally.
