Frequency Therapeutics — Hearing Loss Regeneration

Per this, the company has confirmed their study is impacted by COVID-19 and as a result they will try to get by with less enrollment. Their timeline is still affected, however, and they will update as they know more.
Just finished reading the article, I take this as really good news, or at least the best we can expect.

They basically said that they think they can still hit their key objectives for phase 2a with less enrollment, but can't give an estimate on when results will get out since it is unknown what the future impact of COVID-19 will be.

It seems like, as of now, it hasn't stopped them.

Let's say that your tinnitus comes from your brain, caused by hyperactive neurons, how could regenerating your hair cells undo the problem?
Perhaps the lack of the input of the hair cells create the hyperactivity (think of the non-t person who experiences t in a silent room). Though I don't don't know the source of the stats that ~40% of the people with hearing aid experience reduced tinnitus. This is often pointed to as a real world example of how increased stimuli can decrease perception of tinnitus.

It doesn't make sense that neuroplasticity would only work one way and be "stuck". The brain doesn't have preferences. Reversing the cause should reverse maladaptive plasticity just like mirror therapy treats maladaptive plasticity with phantom limb pain.

Now if tinnitus is from a direct head injury, then yes treating the cochlea won't help.

Was watching some more stuff with Ramachandran and it seems they're also using mirror therapy to try and rehabilitate stroke patients. If that is the case, and if I understand correctly stroke causes some physical damage to the brain, then maybe the brain is capable of rewiring itself around the harm. Been trying for the life of me but can't think up a low-tech mirror box solution for tinnitus.

Ya I think we see this in effect when normies go to concerts and come out with their ears ringing (tinnitus) due to temporary threshold shift. The ringing goes away once their hair cells "recover" over the next 48 hours or so.

I hope that FX-322 can be approved for Expanded Access. This would be fantastic for all of us if it happens. Maybe this could be another question for Frequency Therapeutics during the the Tinnitus Talk Podcast.
FYI expanded access is already legal in the US but Frequency says on their website they aren't scaled up production wise to meet the demand so they aren't granting it at this time.

Whether that could change at any point will be asked, however.

I swear once I get FX-322 I'm going to take care of my ears like they are my children. I'm not going to make the same mistakes.
I was thinking about this today.

I already have some earplugs on my keyring that let sound in up to a certain level; just incase I end up somewhere unexpectedly loud such as really loud bars, or those mariachi bands that will show up at Mexican restaurants and deafen the whole place.

I don't think I'll ever stop listening to music at a somewhat loud level in my car & around the house, it's the only way I really enjoy it...

Firearms are also super present in my family, so I'll be doubling up (earplugs & muffs) any time I shoot in the future.

I know my hearing issues aren't as bad as some others here, but I want this drug to be out so badly. I'm just sick of electric buzzing that never goes away. Everytime I think I'm getting used to it, it comes full circle and bothers me even more.

This COVID-19 stuff makes me nervous too; but a silver lining is that (allegedly) the FDA will be changing their process soon so that drugs can get approved in months, not years.

I doubt any of us fully understood the risk we were taking when we damaged our hearing.

I didn't. :dunno:

Ear ringing sounds trivial, and is trivial, when it goes away.

But the idea that ear ringing could turn on and not turn off, that your ears could scream at you all day and all night, was inconceivable to me before I experienced it.

Hard to imagine any of us making the same mistake again if given a 2nd chance.

I also want to add that I am so excited to be able to use drugs occasionally again.

As of right now, anytime I get stoned or drunk, it seems to make the ringing worse.

Then I get paranoid about it, which ruins the whole experience. Being totally sober at 24 is torture.

I just want to be able to see a movie in a theatre. Take a cute girl out, eat some popcorn. A part of me wants to try with ear plugs, but I'm too scared. (And all the theatres are closed now I guess.)

What do you guys think will be the testing standard for those of us with tinnitus but no hearing loss on a standard audiogram? Will extended audiograms be utilized more?

Seems like there are many of us on here who were told we have "perfect hearing" but we all know about hidden hearing loss.

Haha, best reason I heard so far to get rid of it. Really hardcore hangovers are actually when my tinnitus is the lowest, it's super weird.

I still go to movies (when they aren't closed down) and date girls haha. Personally I am really only concerned with abnormal amounts of noise, such as gunshots or indoor concerts.

I'm not sure what the nature of your hearing problems are, but I try to live my normal life as much as possible. Not doing the things I used to do just makes me focus on the ringing more & then I get depressed as a result.

I do have a feeling that FX-322 will work and will give us a second chance to live our lives in peace without the ringing.
I agree. Even if it doesn't fully resolve our tinnitus, I think for most of us it will reduce it drastically.

Then it is just a matter of waiting for one of the synaptopathy drugs to hit the market for our ears to be fully regenerated (or at least damn close).

Not going to lie it is like my #1 reason for wanting it gone. :beeranimation:

The hangover thing is super weird! Maybe you're focusing so much on the headache that you forget about the tinnitus?

I will actually be microdosing magic mushrooms for a couple of weeks soon.

They've been shown to introduce heightened levels of neuroplasticity, and for some people their tinnitus is gone or significantly lessened by the time they stop using the mushrooms.

I heard Hough Ear Institute is working on a synaptopathy drug or is it Otomony? I'm not sure.

If Strekin AG was able to complete its phase 3 clinical study of STR001 to restore hearing during this critical time (coronavirus), I don't see the need for FX-322 to have any delays as well...

Soon we will see the results of this new drug (STR001) and start growing some hair cells!
