Frequency Therapeutics — Hearing Loss Regeneration

is there any supplement that increases NT-3 in the circulation that you have found?
"In addition to protecting the brain from the development of plaques and tangles, lithium has been shown to repair existing damages brought about by Alzheimer's disease pathogenesis. Lithium ions, for example, encourage the synthesis and release of key neurotrophic factors such as brain-derived neurotrophic factor (BDNF) and neurotrophin-3 (NT-3), which in turn stimulate the growth and repair of neurons. Patients on lithium have been found to have significantly higher gray matter volumes in the brain. One study has even directly demonstrated that damaged nerve cells exposed to lithium respond with increases in dendritic number and length."

Assuming in X years FX-322 is a success, what do you guys believe the estimated cost would be for said treatment?
In the United States, probably in the thousands of dollars or even hundreds of thousands because I assume that a lot of insurance companies will not pay for it.

Yes. Are you?
I would ramp up slow and be cautious and not cavalier with this one, personally.

How exactly lithium works is a bit poorly understood and they aren't even sure (at this point) if it displaces Magnesium (and therefore increases it's bioavailability in other areas) or acts as a cofactor concurrently with magnesium. Either way, Magnesium itself seems to play a role in how lithium works and, on its own, Mag is *very* helpful for all sorts of nerve dysfunction. Would be curious to see how lithium works.

2 weeks until their announcement. Will it be a big nothing burger? Will there be any new information?
Isn't it meant to be an extremely brief presentation? A few minutes? I can't see it mentioning tinnitus anyway. What could they say other than they're on track to positive results? Enough to keep investors happy perhaps.

Yeah, that is what I expect but I can always be optimistic.

I follow this thread and a sudden question popped into my mind, how will the FX-322 treatment be delivered / offered once it's out?

Will we get it through a doctor? Will we get it through a pill (I'm pretty sure it's an injection in the ear or something though, so I don't know)? I really want to know because of how much it could help people. It could even help me, or maybe even if you have tinnitus with hearing loss.

Sorry if this is a pointless question to ask, just popped in my head and decided to ask it while I still remembered.

Injected through the ear.

Sorry if this question has already been asked, but how does this treatment get to our inner ear? I remember reading a paper by Stanford a few years ago which stated that as long as we cannot get through the cochlea, we cannot reach the inner ear for regenerative purposes.

How have they solved this?

In the middle ear there is a membrane between it and the inner ear called the round window membrane and small enough particles can diffuse through it.

How have they solved this?
By developing a solution with the right chemical properties to permeate through the round window membrane. Also, their drug turns from a liquid to a gel after injection into the middle ear. The gel provides longer contact duration.

From https://www.ncbi.nlm.nih.gov/pmc/articles/PMC6557301/

"The round window membrane (RWM) is the most accessible of these structures; absorption of drugs through the RWM is dependent upon permeability properties and the contact duration of the applied drug substances."

Thats's amazing! Like all of you I'm hoping for positive results.

Any idea why Frequency Therapeutics can do this while researchers at Stanford are puzzled?
Who's to say they're puzzled? Maybe they are working on their own version and just aren't as far along (or are being quieter about it).

Is FX-322 really proven to be effective? Or is it just a claim made by Frequency Therapeutics?
After the AM-101,I am very cautious about any potential therapy.
The ruin of hope is most terrible.

A question like that is guaranteed to sow discord around here. Not even Frequency Therapeutics would claim it's proven, but some sophisticated investors have bet money on it. It's looking hopeful. We'll know more within a few weeks. 'Cautiously optimistic' is what I aim for.

This is what I worry about.

A report driven by investors may be distorted. Sophisticated investors don't mean professionals. They care about the company value more than the actual result.

Hope I was just influenced by the AM-101 event, and I found that the AM-101 is still in development.

The difference here is that increased word scores and improved audiograms couldn't be faked without full on fraud. Not that fraud isn't theoretically possible, but I see too many reputable researchers behind Frequency Therapeutics for this to likely be the case.

Not to mention the US government is one of the investors so you have government and private investors and their reputations behind this as well as the backing of many high level scientists.

Tinnitus scores and THI, on the other hand, are harder to quantify as objectively. And didn't people lie about how long they had tinnitus to be part of Auris' study? Wasn't that part of the problem or did I invent reading that in my head since I can't seem to locate that now?

Charles Liberman thinks we have a solution to cochlear loss coming up soon, that's good enough for me.

Though in my case, I'm now holding out for Autifony to find a good KV3.1 channel opening drug because I think that's much of my problem and it might be an even more difficult one to solve :(.

Frequency Therapeutics isn't publicly listed. There's no share price to influence. One of their 'investors' is a pharmaceutical company who bought in (for up to 620 million USD as milestones are met). I'm guessing that this means that no more investors are required. There's no payoff for anyone now unless it works.
