I Was a Guinea Pig in the Autifony QUIET-1 Study, Great Results

Really happy and positive news indeed for the T sufferers all over the world. Remember this positive result is from a well documented trial and not from some patch maker who market their stuff with Facebook and celebrities or some miracle cures without any proof. This is for real, folks. I am so happy to hear about this news for the T community and all those who need their T cured or treated, not just habituated. Let's hope it will be available soon to bring the T curse to an end. Remember this is only the 1st drug from them Quiet-1, so future improvement on the drug is possible with -2 & -3 etc, and newer drugs can come out to the market from other major pharma once the direction of the cure is established by this drug. It will come a time when a final wonder drug will be developed. If this version can reduce 75% and need to be taken continually, this is as good as it can be. People have to take drugs continually for blood pressure, for diabetes etc. etc. So no big deal as long as the side effects are minimal.

Can you imagine a day when ENTs of the world won't have to put more stress and suffering to their new desperate T patients, by putting the nail to the 'coffin of T curse' when saying to you that your unbearable T shrill is there for life and you have to live with it. Gosh!!! How many times you have read new members saying this 'verdict' instantly sinks them into despair and panic attack. When more of these medically proven and tested new drugs for T are available, hopefully, the ENTs will prescribe these instead of their dreaded 'verdict of death'. LOL. When the new T patients know there is something to help reduced the volume, the major part of T suffering, the mental 'hell' will be over and T will have very little power to rob you of your life. Hurray! The 'hell' with T. Amen.

I just can't wait for people to be told by their doctor, yes, we have a treatment, not be told to just live it. I can't believe it's taken this amount of time...But for new sufferers I'm glad they won't ever be told to just be told "just live with it". I was looking up KV3.1 modulators and in 2003 they knew the role they played in tinnitus...It's pathetic that in 2015 we have no drug available on the market to treat this condition.
 
Finally, a point that I haven't seen made elsewhere. This is very much a first in class treatment, and could be the first to market treatment for tinnitus. The importance of this should not b e understated. Firstly it will establish the market for treatment (GSK and Pfizer et al. do not target blue sky projects) and hence lead to commercial scale investment in tinnitus RnD. Secondly, it will prove that potassium channel blockers are a viable treatment path. The combination of the two will finally result in likely drastic improvements in the pharmacological treatments (think the next iteration of trobalt). TLDR; If AUT'63 proves successful this will rapidly lead to a whole family of likely much improved treatments with far better outcomes even then the original drug.

The other thing of note is that tinnitus is thought to share many things with various other "untreatable" conditions, including CPRS and HPPD. So, if this class of drugs does indeed have efficacy for tinnitus, then it might open the door to treatments for those conditions, too.

This is great news especially for those of us who have catastrophic intractable T. I am sure the less happy of us would tolerate the reflux issue to gain some peace. Thanks for sharing your experience.
 
On a brief pharmacological note, in this source from Autifony I read that the dose for AUT3 was 30 mg/kg on Long Evans rats which have their weight specified in scientific sources.

The primary outcome measure on the Autifony wesbsite states

"Primary Outcome Measures:
  • Change in subjective Tinnitus after 4 weeks treatment [ Time Frame: 28 days ] [ Designated as safety issue: No ]
    To compare the change from baseline (D1 to D28) of the Tinnitus Functional Index overall score between AUT00063 (800 mg) and placecebo."

After doing the math, it seems to me that Phase 2 stood way on the safe side and the administered dose was at a much lower concentration in humans. Perhaps to stay consistent with the Primary outcome measure

So definitely now that we know there is a therapeutic effect, the dosage shall be much improved.

Great point i have 5 different sounds some 3 stay all day 2 come and go.. Im pretty sure it should work for everything not just one tonal t, as everyone has different sounds..
 
I don't agree with the negative comments on AM 101. I have had 2 x sets of injections and had good success with a 30 - 40% permanent decrease in volume (not temporary like Autifony). I have two more sets of injections to attend - first one in July 2015 second set I presume later in the Autumn. I think the people who are writing negative comments have either not had two sets of injections themselves (because first could have been placebo) or they are too scared go through the process so are latching onto anything negative. I don't expect to achieve silence but any improvement is an improvement on lifestyle.

Hopefully, YES! If his drug really helps I really wish for him and his team to get filthy rich, have alle the best sex they can hope for and live a healty beatuful life to they are 150 years old. I for one will send him flowers on his birthday for the rest of my life with a card saying I will never forget what you have done! Anyway, this is what I wish for!

@Mrs D apologies if this was already asked but did you have hyperacusis or any sensitivity to sounds? Did that also improve while on the trial?

Retigabine and Maxipost are two different medicines and they discovered that when taken after noise exposure the one or the other would obstruct a high tone tinnitus or a low tone tinnitus to appear ... RTG was low tone T ... maxipost the higher. I do think that these potassium regulators will have effect on both but maybe more storngly on one or the other ... still very mysterious

I wonder what would happen if you combine Autifony's drug with trobalt?

That is actually really good news as it seems that these injections do work on slightly older T as you have had yours since November 13.......brilliant and thanks for that post...........

I have a question also: Would you say that the T. came back gradually, during those three weeks, after the last dose or did it hit back as sudden as the first time you got it?
