So, yesterday I had some board games at home and I used a vape from one of my friends. It was pretty strong on synthetic nicotine (50 mg/ml) and I hit it 4 times. Also, I took 2 glasses of wine.

Can this kind of nicotine destroy the inner hair cells instantly when you have too much in one day?

My tinnitus was noticeably louder when I went to sleep and still today it's a bit higher than the baseline.

Not sure if my right ear feels muffled or if I am tripping in obsessing... Is such a thing possible?

Your thoughts on this are welcome.

Caffeine, salt, sugar all spike my tinnitus. So does stress, lack of sleep, worry. Every spike from those substances and some offending supplements all return to baseline for me. I just drank a Coke because I have a lot of work today and my tinnitus is screaming. It will be back to baseline by tomorrow. I don't smoke or consume nicotine so I don't know anything about that particular substance.

I don't have experience from vaping. But from my experience drinking alcohol makes my tinnitus worse. I have always been thinking it was dehydration but I often also seem to get in a bad mood the night and day after drinking which doesn't help on my tinnitus either :)

Drinking only gives a very brief relief from tinnitus for me because it distracts me and I have found out I can get that distraction with non-alcoholic drinks and fun activities too. It's been a long time since I have played board games and I should go back into that (I play computer and video games mostly).

No, these kind of spikes are temporary.
