i worry xenforo threads may have a limit
we'd be justified to make an MPP 2.0

it's an addon for the forum, that's how these forums work through user downloaded addons some free, some you have to pay. A good programmer might be able to make their own. I guess.

but yeah it's possible MPP might suffer this fate in the near future.

Professors, PHD's and respectable scientist helping our cause like Liberman, Shore and Schaette and bio companies like Frequency-tx have have most likely browsed MPP. bio companies keep track of their online presence and universities, the BTA monitors this forum.

We're probably not much interest, but I'm sure they have all checked into MPP a few times especially with my amateur deciphering of research papers which I was told to stop after being scolded for over simplifying complex research even though I had well intentions, acting like Wannabe PHD Otologist.

I just don't know how they feel or would feel if there research, bio companies became a meme.

Scientist outside of NeilDegrasse Tyson, Elon Musk and Bill Nye aren't use to that kind of publicity. so i don't know if MPP making hearing loss/tinnitus research into a meme is a good thing or not.

Once again it adds insult to there work, because we are not mentally healthy people, I'm a barely employed graphic artist that life was ruined by hearing damage, I openly advocate radical left wing nihilist ideas on this forum while still having an edgy offensive sense of humor, it's even true I was still failing at life long before hearing damage, just a part time job and starting classes several years behind most people my age because pre existing mental health issues.

my life was pathetic before but atleast I had some things going and a potential future if i worked hard. i loved my job, volunteer work, gym and was so excited to be in school for just a short period of time before my life ruined, I am literally haunted that i threw my life away.

threefirefour, johnadams and I are probably viewed as jestures to them, probably on lunchbreak they check tinnitustalk on their phones cringing at us. however it's only a matter of time before Jastreboff takes legal action.

Who cares? They can cringe all they want. They dont own knowledge. History is full of examples of people without degrees or formal educations changing the world. Like Gregor Mendel and Michael Farady. Last time I checked, it wasnt FrequencyTX that discovered hair cell regeneration, they are just the ones testing it in humans. Perhaps your efforts would be better utilized doing like me and polishsoldier87 and trying to find a way to cure ourselves instead of denegrading our lack of phD's.

Dude I seriously love the way you think. I've been trying to get onboard this choo choo and read these PubMed/studies and understand WTF is going on. There HAS to be a way we can solve this shit.

Ly411575 and DAPT arent the only substances that regenerate hair cells.
ALSO ly411575 worked orally it just had toxic side effects.
Surely these arent the only substances that can achieve this. We need to start identifying the genetic mechanism behind this, which is probably over expression of the gene atoh1. A good thing to do would be to find out what other mechanisms cause over expression of atoh1. And whether or not some plant or plant extract can achieve this.
Celestrol so far seems like a potential candidate but the safety is suspect.

Celastrol enhances Atoh1 expression in inner ear stem cells and promotes their differentiation into functional auditory neuronal-like cells

During inner ear formation, the Atoh1-positive cells are committed to generate sensory hair cells and indispensable for their development, survival, differentiation and maturation. The ectopic introduction of Atoh1 led to formation of supernumerary cochlear sensory hair cells and aberrant overexpression of Atoh1 stimulated trans-differentiation of supporting cells to hair cells.

Here we presented evidences that Celastrol significantly improved inner ear stem cells viability and proliferation. Moreover, Celastrol stimulated neuronal-like cells differentiation and excitability, which might be mediated by up-regulation of the master transcription factor Atoh1. Our results warranted further investigations into the potential therapeutic value of Celastrol in disease model and clinic trial.

On the other hand, green tea/EGCG overdose cause adverse complications including acute liver failure/hepatotoxicity via alterations in liver enzymes. An experiment based on rat model revealed that oral dose delivering 2000 mg EGCG preparation/kg was lethal to rats; but dose with 200 mg EGCG/kg induced no toxicity [81].

Ok so it looks like 200/kg EGCG is safe according to that paper. Thats 13.6 grams for a 150 lb person. That seems like alot. The ly411575 is a gamma secretase inhibitor and was fed to the rats at 50/mg per kg. That's 3.5 grams for a 150 lb person. So I probably dont need 13.5 grams of egcg. I guess I'll start at 1 gram per day and see how I react. Hope I dont die.

EGCG is a gamma secretase inhibitor
Ly411575 can restore hair cells
EGCG (according to that paper) can restore hair cells.
Ly411575 can achieve this orally
EGCG can be taken orally, allegedly in high doses.

Next question is: can EGCG, taken orally, restore hair cells?

Is there any decent chance that hearing regeneration will also cure my ear pain, ear fullness and overall hearing distortion?

Seriously, with ENTs still stuck in the dark ages with their 8 khz audiograms I don't even know why I'm still around. Somebody give me hope? But don't lie to me.

possibly, but I'm silent on the issue because we don't know and I don't want to make assumptions.

there are still suppose to be different types of hyperacusis that need to be classified.

I disagree, they did a lot of hardwork and schooling to get a PH.D and the long ass sciencey papers that I amateurishly deceipher shows that.

I'm not saying they arent smart and capable. I am saying we are potentially capable too. We dont have multi million dollar labs and access to genetically modified rodents, but they do and publish their results. The difference is that they have constraints related to planning and FDA regulations and we dont. It's as if there is an untapped synergy between people like them and us.
