So correct me if I'm wrong but it looks like their hair cell regeneration technology is based on injecting siRNA into the eardrum to target the Notch signaling pathway.
Notch signaling is the key. I've known this since my curcumin experiments.
This is it, the key to regenerating hair cells. Now we can only hope that restoring hair cells will correct hyperacusis and tinnitus. Also we can only hope that THIS HAPPENS QUICKLY!!!!!
Recap:
The Hough Ear pill is Disufenton/NAC.
And the injection is based on Notch signaling from siRNA injected into the middle ear.
I've just searched the FDA trials site for siRNA + hearing loss and couldn't find anything which is confusing because Dr. Home Boy said he expects it to be approved by 2020.
We need to figure out something to make our BLBs permeable to something else that affects the notch1 signalling pathway.
Curcumin is a potent notch1 inhibitor, but I took enough of that stuff to make curry for the entire Indian subcontinent and it didn't do anything. Maybe if we paired it with something that increases permeability of the BLB like Mannitol then it would enter the cochlea better.
Our bloodstream is separated from the fluids in the cochlea by the BLB (blood labyrinth barrier).
Mannitol makes BLB more permeable (permeable means stuff can go through it).
Curcumin as a Notch1 inhibitor
"An oral glucose tolerance test and an intraperitoneal insulin tolerance test revealed that insulin resistance was reduced (P<0.05 or P<0.01) and tissue section analysis revealed that hepatosteatosis was attenuated following treatment with curcumin. Furthermore, the protein expression of Notch-1 and its downstream target Hes-1 were suppressed. "

Interestingly, Curcumin acts on the Hes-1 gene and so does the siRNA injection that Hough Ear is working on.
I'm suddenly very interested in curcumin again.
You can buy Mannitol. I don't know if this would have the same effect on the BLB as the medical grade that's injected into the blood or what the proper dosing would be or of this is even the same thing at all.

Actually, I think Manitol and Mannitol are in fact the same thing.
