Those forums are full of hypochondriac powder puffs. Some of them complain about the temperature of their bath water. That their glass of wine causes headache. Their audiologist is their best friend...

You'll get blasted by some control freak moderator on a power trip. Two or three at a time. They defend everything like corporate shills.

To the point I wonder if they're promotional sites defended by shill moderators and shill admin.

Smart people realizing and leveraging fβ as free advertising to large targeted market segments.

I went a full page of replies with an admin who jumped me before he said "okay, go for it", regarding promoting a push for a cure.

Then a week later they're back on you.

Like any open online community the majority of users are non-registered lurkers.

There are probably a dozen times more lurkers then active users at any moment, Steve and Markku are the only ones that would know the exact stats, but registered users here are probably 95%-99% dormant accounts.

Those people on Facebook can be awful, I tried helping someone by posting a link to the mutebutton q&a to give them hope and the admin immediately banned me
 
Notch inhibitor. Gamma secretase. Thats what triggers lgr5+ cells to grow. I read elsewhere there needs to beta catenin present for the cells to differentiate into hair cells.

Jeez don't take a gamma sectretase inhibitor. The clinical trials for these for Alzheimer's partly failed due to severe side effects. Gamma secretase has loads of substrates in the body

They were taking them in higher doses and orally, local injection into the middle ear like fx322 is much safer. They already stated that after the administration of it they checked the patients blood and there wasnt any.

Yeah youre right. But when people get tinnitus, they go to r/tinnitus or TT, usually for casual browsing to see if there's any treatments or whatever. So even though taking the tinnitus facebook is a good idea, I think the main priorities are r/tinnitus and maintaining TT. If we have both of those, then the new sufferers will learn that treatment needs to come first. Without new blood, the facebook will either be forced to accept that treatment is the new king and habituation is dead, or they will stop getting new members, shrivel up, and just become an obsolete habituation echo chamber like TSMB.
 
i believe the early experiments involving rats was oral administration in very high doses, like 50mg per kg of body mass, for several days. it discolored their skin, made their hair fall out, and gave them tumors, but their hearing came back.
Basically gamma secretase interacts with Notch and Notch is involved in sooo much stuff, hence side effects skin cancer etc in human trials. You sound like you know enough to realise that proteins with many substrates make bad drug targets unless they can be targeted in some specific way. But my knowledge not good enough to know how this could work in the ear, whether there would still be any potential for at least some systemic effects.

disruptions and irregularities in the wnt/beta-catenin signaling pathway are implicated in many types of cancers. notch receptors as well as you have stated. that was my goal with taking lots of curcumin, is that is a notch1 inhibitor. it also does other things that cancel out it's ability to cause over expression of atonal homolog 1 which is implicated in causing lgr5+ progenitor cells to proliferate and regenerate. :(

Notch1 inhibition increases histone acetylation which increases atonal homolog 1 which initiates hair cell regeneration.

Curcumin is a notch1 inhibitor but it also stops histone acetylation so it was like putting on the gas and the brakes at the same time.

this is why they are injecting fx-322 into the eardrum.
 
not being able to enjoy music the way i did geniunely makes me want to kill myself to be honest
hearing loss and tinnitus are hell to deal with but knowing the slighest nonsensical sound can make it much worse is like living on death row
waiting for the day you'll feel comfortable enough to enjoy yourself and just kill yourself with music

@Manny you ever heard of pokemon showdown? it's a website where you battle other people, it's pretty cool
I have, and I once tried it briefly! The problem is while I love Pokemon, I seriously suck at competitive battling. To be fair, I never put effort into it.
I'm good at knowing what will be super-effective (in gens 1-3) but that's about it. My "strategy" is basically just try to get max-stat Pokemon and use the highest power and super-effective moves, LMAO. Works just fine for in-game, but against competitive battlers I'm a total goner.
